The fundamental theory behind the use of a throttle body spacer is to lengthen the intake runner to shift the torque curve lower in the RPM range. An engine with a torque peak high in the RPM range can benefit from a throttle body spacer. If an engine already has its torque peak low in the RPM range, or has an almost flat torque curve, there is no benefit. All of the 4.3L v6 engines fall into the later category.

I had ones CNC cut for my old 3.5L Intrepid and gained a solid 2mpg with them. This engine had very little low RPM torque and showed decent benefits from the addition of the spacers.
