I have my 95 Blazer with CMFI and I met someone that has the same 4.3L in a 2001 Chevy S10 pickup. He has had to have his motor replaced three times and so he has a spare electronic fuel system, with regulator, he wants to give me. Can I convert to this system without issue?

Your 95 has CPI, not CMFI. CPI stands for central port injection which has only one electronic injector that fires in a batch mode feeding 6 individual pressure actuated poppet nozzles located in the intake runners ahead of the valves. 96 was the first year for the CSFI (central sequential fuel injection) which had 6 individual electronic injectors fired sequentially housed in a central injector pack with each individual injector feeding a pressure actuated poppet nozzle located in the intake runner. Somewhere in mid-01 was the CMFI update that moved the electronic injectors out to the intake runner location replacing the often problematic poppet nozzles.
As far as what you can do... Just make sure that everything is working as it should. Not much else can be done to the CPI engines.

As Swartllk described, the entire powertrain control system is incompatible between 1995 and 1996. So no switching of computer or computer and wiring either. Honestly, I doubt if switching a properly working system to the newer design would get you any additional fuel mileage. Check for sticking calipers (brake drag is really a fuel mileage killer) and drive with a light foot. Myself, I get about 13-14 mpg on average with my 2001 4WD running A/C and combo driving 1/2 at 45mph in country + stop/go city. 17mpg on Interstate at 70mph with A/C. I have a 2004 Chevy Avalanche (think pickup made from a Suburban) that does better. Blazers are definitely not fuel efficient vehicles and probably why the platform originally fell. Personally I think the difference in price of an ugly new frameless SUV will buy a heck of a lot of fuel.
