The biggest benefit to using a gasket is that you can torque down the cover bolts, fill the diff, and drive away. With RTV, you have to wait for it to set up before you fill it and really shouldn't drive it until it is fully cured.

For diffs, I wouldn't use anything but synthetics! GM has a very good synthetic 75w90 that is the factory fill in the newer trucks.
