Default What to tune?

I recently acquired a Jet DST/TunerCats type device and software. I have updated the computer(16244210) to the latest OS and all modules per the GM vin software check website.

I have also done these modifications to my calibration:
-Applied "Speed Calibration" for 4.10 gears and 32.7" tires as that is the actual BFG AT KO 33" size. (Modifies transmission tables.)
-Set "Fuel Cutoff/Resume Vehicle Speed" to 255mph and 252mph respectively. (Removes speed governor.)
-Set "Torque Management Spark Retard Limit vs RPM" table to all 0s. (Removes torque management.)

So... My question is; what can I do to actually get more performance/torque or mpg out of my engine/transmission?

Physical engine/trans mods: (Engine is L35 4.3L, Transmission is 4L60E)
-3" CAI
-2.5" Dynomax cat-back exhaust
-External transmission cooler and filter (adds 1.5-2qts to system)
-Steel hard-parts in transmission (HD components)

P.S. I have a couple VCMs so my plan is to flash one with performance mods and paint it red, and paint one blue and tweak it for mpg. That way I can swap on trips and not waist all my trail gas on the highway!
